What is Semax?

Semax is a synthetic heptapeptide derived from the N-terminal fragment of adrenocorticotropic hormone (ACTH 4-7). It carries a C-terminal Pro-Gly-Pro extension. Semax was originally developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. It was designed as a tool for investigating neuropeptide signaling pathways.

The ACTH(4-7) fragment (Met-Glu-His-Phe) is a sequence identified in early neuropeptide research. It retains neurotropic activity without the adrenocorticotropic effects of full-length ACTH. The Pro-Gly-Pro C-terminal extension was engineered to enhance metabolic stability against peptidase degradation. This extends the compound’s functional duration in experimental systems.

Semax is one of the most widely referenced synthetic neuropeptides in neurotrophin signaling and neuroprotection research. It has a substantial body of published literature from Russian and international research groups.

Supplied exclusively for in-vitro and ex-vivo research by qualified laboratories. Not intended for human or animal administration.

Mechanistic Overview

Neurotrophin Pathway Research
Published research has examined Semax in models of neurotrophin signaling. The primary pathways studied are BDNF (brain-derived neurotrophic factor) and NGF (nerve growth factor) expression. In vitro studies have characterized effects on neurotrophin mRNA expression levels and downstream TrkB receptor signaling in neuronal culture models.

Melanocortin System Interaction
As a derivative of the ACTH(4-7) sequence, Semax has been investigated for interactions with melanocortin receptor subtypes in CNS tissue models. Research has examined its binding profile across MC3R and MC4R. Studies have also characterized the relationship between melanocortin receptor activation and neuroplasticity-associated signaling cascades.

Neuroprotection Models
Semax has been studied in experimental oxidative stress, excitotoxicity, and ischemia-reperfusion models. Published findings describe effects on inflammatory cytokine expression profiles. Research has also characterized antioxidant enzyme pathway modulation under controlled experimental conditions.

Reconstitution & Storage

Lyophilized Semax is stable at -20°C for long-term storage. Reconstitution for in-vitro work is typically performed using bacteriostatic water or sterile saline. Reconstituted aliquots are generally stored at 2-8°C for short-term use. For extended storage, aliquot and freeze at -20°C to preserve peptide integrity. Researchers should follow institutional SOPs for handling lyophilized peptides.
